Grandevity

Grandevity is our cutting-edge Longevity supplement. Once daily, Grandevity activates lonegvity genes! Nrf2
 and Longevity

Nrf2 is a protein in all the cells in the body.  When activated, it relocates from inside the cell membrane to the nucleus of the cell.  There, it attaches to a specific segment of DNA called the Antioxidant Response Element.  Genes downstream on the DNA are then upregulated or downregulated.  Older Nrf2 technology was known to up-regulate antioxidant enzymes like Superoxide Dismutase, Catalase, and Glutathione.  Additionally, there was upregulation of other beneficial genes and downregulation of the production of some harmful proteins such as NF-KB.  With newer research, it has been determined that some of the older Nrf2 technologies may cause some detrimental effects on the cells. Enter Grandevity!

Based on the extensive amount of new, published scientific literature, perhaps the single most significant new area is the “shutdown” part of Nrf2 activation;  Nrf2 that is activated by dietary signals remains in the nucleus a little longer and induces more expression of protective genes. Most biochemical systems, in fact, have these types of “on” and “off” pathways running concurrently but at different rates to provide quick response time. 

 

Grandevity, makes use of this expanding knowledge base of Nrf2 activation.  We now know that Nrf2 activation consists not only of the Nrf2 transcription factor being released from its inhibitor, migrating into the cell nucleus, and binding to specific DNA sequences that promote cytoprotective gene expression but also of modulating the rate at which Nrf2 is removed from the nucleus. What is Oxidative Stress?

Oxidative stress is rusting of the body from the inside out.  A rotting apple is a perfect example of oxidative stress. What causes oxidative stress? Oxidative stress, or oxidation,  is a disturbance in the balance between the production of reactive oxygen species and the antioxidant defenses.  The foods we eat get "burned" as fuel.  There is waste or exhaust associated with this process of making energy for the cells.  This "exhaust" or "waste" is the free radicals.  These free radicals can damage any part of the cells they contact.  What is ROS? Reactive oxygen species, aka free radicals. What are free radicals?  Free radicals are toxic reactive molecules that will damage any part of the cell they bump into. Our bodies have a built-in natural defense system.  We produce anti-oxidant enzymes which can neutralize those free radicals before the can damage our cells. The Nrf2 pathway senses the need for these antioxidant enzymes and regulates their production to maintain this delicate balance. As we age we produce fewer of these antioxidant enzymes such as superoxide dismutase, catalase, glutathione and others.  Nrf2 production declines with age as well.  Multiple health conditions and diseases are a direct result of oxidative stress.  Infections, allergies, and autoimmune diseases activate our immune cells, which produce free radicals in an attempt to kill germs that the immune system assumes to be present.  Unfortunately, our otherwise healthy cells get caught in the cross-fire and sustain collateral damage that we see and feel as inflammation.  Other major diseases associated with aging, such as heart disease, stroke, cancer, and neurodegenerative conditions such as Alzheimer’s disease also increase production of free radicals, creating oxidative stress, and inflammation.

Why is Nrf2 Important?

Our DNA encodes about 20,000 genes, each representing a “blueprint” for the production of a protein or enzyme necessary for a healthy existence.  Each of these “blueprints” requires a regulating control called a “promoter” that determines precisely how much of each product is produced, and under what circumstances.  By binding to one specific type of these switch-like promoter regions called the “Antioxidant Response Element (ARE)”  Nrf2 regulates hundreds of different genes that allow cells to survive under stressful conditions.   These genes produce antioxidant enzymes that protect the cells by neutralizing free radicals and by helping to repair the damage they have caused.

How can Nrf2 be Activated?

The Nrf2 protein has the ability to control the expression of genes (epigenetics). The Nrf2 pathway senses and responds to changes in oxidative balance.  Nrf2 can be activated when an imbalance is detected.  It can then “switch on” or “turn off” the genes it controls (often termed survival genes) to produce the protected state within the cell.  Many of the chemical compounds that are Nrf2 activators are produced by certain plants, crude extracts of which were known to our ancestors centuries ago in traditional Chinese, Ayurveda, and Native American medicines.  These plant-based compounds (phytochemicals) appear to be just as potent or more potent than, pharmaceutical Nrf2 products products that are beginning to emerge.  The power of Grandevity is in the multitude of positive effects it has on multiple longevity genes in addition to its ability to significantly reduce oxidative stress. Why You Should Activate Nrf2 ?

Why should we be concerned with supplements or therapeutics that can adjust the Nrf2 pathway? Isn’t it, after all, part of a very large network of interactive and self-regulating pathways that automatically adjust to achieve proper balance based on our metabolism?  In a young healthy person, this appears to be the case.  After a person is past their reproductive years, however, things begin to change, as Nrf2-activation capabilities decline with age.  As harsh as it may sound, after we have passed on our DNA and reared our children we have mostly served our purpose as far as Mother Nature is concerned. Many studies have shown that the concentration of Nrf2 in cells declines as we age.  Old age brings with it increased markers of oxidative stress and an assortment of “age-related diseases” such as atherosclerosis and cardiovascular disease, cancer, arthritis, cataracts, osteoporosis, type 2 diabetes, hypertension, Alzheimer's and Parkinson’s diseases, and many others.  All of these diseases have at their core increased oxidative stress.  By gently stimulating our aging cells’ ability to activate and modulate Nrf2, we aim to restore and enhance our body’s own ability to counteract the effects of oxidative stress so we can live to a 'Great Age'.

 

TBARS (thiobarbituric acid)—which is a measure of oxidative stress declines with Nrf2 activation. Elevated TBARS have been associated with a significantly increased risk of cardiac events. Explosion in Nrf2 Research

In 2004, there were about 200 papers in the literature on Nrf2 and we were just beginning to paint the picture of Nrf2 as the “master regulator” of the antioxidant response in mammals.  Now, in 2023 there are more than 25,000 academic studies have been published, and the title of “master regulator” has proven to be well-deserved.  Nrf2, in fact, regulates thousands of genes that have nothing to do with antioxidant enzymes per se, but rather provide protection from a broader range of stress-related events that are encountered by cells, organs, and organisms, under both normal and pathological circumstances.  We are now in a much better position to redesign, from the ground up, a new Nrf2 activating dietary supplement based on this massive amount of information learned from published academic research.

As recently as 2007  Nrf2 activation was thought of in rather simplistic terms—pretty much a one-step process of activating a specific kinase that phosphorylated Nrf2, releasing it to enter the nucleus.  The years since then have produced thousands of studies shedding much more light on the intricacies of the pathway.  Many of the newly discovered steps in the Nrf2 pathway help us understand the enormity of what this pathway can do in our bodies.  Grandevity, supports longevity genes. Older Nrf2 activators do not activate these longevity genes.

 

Biochemical pathways must be responsive to changing needs and able to sense changing conditions to maintain proper balance. Our current view of the Nrf2 pathway is much more complicated than was appreciated using this knowledge to our advantage; which was a decade ago. Grandevity supports longevity and Nrf2 activation.
